Background :Hypoxia-inducible factor 1-alpha: Functions as a master transcriptional regulator of the adaptive response to hypoxia . Under hypoxic conditions, activates the transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, HILPDA, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia . Plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease . Heterodimerizes with ARNT; heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters . Activation requires recruitment of transcriptional coactivators such as CREBBP and EP300. Activity is enhanced by interaction with NCOA1 and/or NCOA2. Western blot analysis of Galectin 3 expression in HEK293T (A), Hela (B), mouse kidney (C) whole cell lysates.

Immunohistochemical analysis of Galectin 3 staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
