Background :The mammalian splicing factor SF3a contains three primary subunits: SF3A1 (SF3a120), SF3A2 (SF3a66), and SF3A3 (SF3a60), which, along with SF3b, activate the U2 snRNP to form the pre-mRNA splicing complex. SF3A2 and SF3A3 interact with SF3A1, but not with each other, and SF3A3 plays a major role in recruiting SF3A1 to the complex. Each protein in the heterotrimer has been shown to be essential for mRNA splicing and cell viability. Notably, SF3a plays a role in growth factor signaling as SF3A1 can bind to ERα when phosphorylated at Ser118 to affect downstream splicing. SF3a also plays a role in the innate immune response, promoting inflammation by splicing the TLR adaptor Myd88. Furthermore, SF3a regulates several genes involved in innate immune signaling pathways in macrophages versus other pathways.
